摘要

Abstract

Objective To study on purifying the total alkaloids in Atalantia buxifolia with macroporous resin. Methods The content of total alkaloids were determined by acid dye-colorimetry. The content of total alkaloids as index, the type of macroporous resin, sample volume, concentration, pH and volume of elution solvent were optimized by single-factor experiments. Results Optimal purification process conditions were as follows:optimal macroporous resin type AB-8, the concentration of sample solution 0.14 g·mL-1 with pH as 10.0, maximum sample volume 5 BV, washed by 9 BV purified water and 10 BV 95%ethanol, collected eluent of 95%ethanol. The average transfer rate of total alkaloids was 83.61%, and the adsorption and desorption rate were 89. 30% and 93. 64%, respectively. The RSD was 0. 30%. Conclusion The total alkaloids in Atalantia buxifolia can be purified effectively by AB-8 macroporous resin with good reproducibility.
