Abstract
Objective To study the influence of edaravone on the proliferation and apoptosis of human ovarian cancer resistant cell lines A2780CP and that on sensitivity to cisplatin .Methods Peak plasma concentrations of cisplatin and edaravone were taken as the basis of combination therapy, and the study included five groups: group of parental A2780 cells treated by cisplatin, group of A2780CP cells treated by cisplatin, group of A2780CP cells treated by edaravone , group of A2780CP cells treated by cisplatin combined with edaravone , and control group .After incubation for certain period the cells relative activity was detected by MTT assay and the apoptosis of cells was analyzed by flow cytometry .Results A2780CP cells treated by cisplatin combined with edaravone grew more slowly than A 2780CP cells treated by cisplatin alone, but the sensitivity on cisplatin increased significantly (P=0.016<0.05).Annexin V-FITC/PI analysis showed that A2780CP cells’ apoptosis could be significantly improved (P=0.037<0.05) with the combining treatment of cisplatin and edaravone after 24 hours.Conclusion Edaravone can improve the sensitivity of A 2780CP on cisplatin significantly , and it has certain reverse effect on cisplatin resistance .关键词
