Abstract
Objective: To investigate the effect of thymic stromal lymphopoietin (TSLP) on the differentiation of intrathymic CD4+CD25- T cells to CD4+CD25+Foxp3+ regulatory T cells in myasthenia gravis (MG) patients with abnormal thymus gland. Methods: Intrathymic CD4+CD25-T cells and dendritic cells isolated by immunomagnetic beads on MiniMACS device were co-cultured with different concentrations of TSLP (0.1, 1, 10, 20 ng/mL) for 7 days. The ratio of CD4+CD25+Foxp3+Treg/CD4+T cells was tested by flow cytometry. The expression of Foxp3 mRNA and protein was measured by RT-PCR and Western blot respectively. The concentration of serum IL-10 was detected by ELISA. Results: The ratio of CD4+CD25+Foxp3+Treg/CD4+T cell, the expression of Foxp3 mRNA and protein, and the level of IL-10 was increased significantly in groups with 10 ng/mL and 20 ng/mL TSLP compared with those in the PBS control group (P<0.05). Conclusion: TSLP may induce the differentiation of intrathymic CD4+CD25 -T cells to CD4+CD25+Foxp3+regulatory T cells in MG patients with abnormal thymus gland and up-regulate the expression of Foxp3.关键词
