Glycerol as a Carbon Source to Produce Epoxide Hydrolase by High Cell Density Fed-Batch Cultivation

Abstract
Epoxide hydrolase is an important industrial enzyme which can stereo-selectively transform racemic epoxide into enantiomeric vicinal diol. In this study, the growth of E. coli Rosetta (DE3)/pET28(a)-CESH on different concentrations of glycerol was studied in shaking flask. Induction time, IPTG concentration and culture temperature after induction were optimized to enhance the production of CESH. The results reveal that in a 15 L fermentor, the fed-batch fermentation process can reduce the fermentation time by 10 hours. Compared with the batch fermentation process, the CESH productivity was increased by 33.3%during the whole process. These results indicate that the new process is suitable for industry application.关键词
