BCL2（Ala43Thr）多态对前列腺癌PC3细胞迁移的影响
THE POLYMORPHISM OF BCL2 ALA43THR EFFECTS ON THE PROSTATE CANCER PC3 CELL MIGRATION

Abstract
Objective:This study aims to research the influence of BCL2 (Ala43Thr)polymorphism on the migration in the prostate cancer PC3 cell line.Methods:PC3 was selected as the obj ective cell line.This ex-periment was divided into 4 groups,(1 )CON:control group,normal target cells,without any virus infec-tion;(2)NC:negative Control,normal target cells,infected with negative control virus;(3)OE-WT:nor-mal target cells,infected with wild type virus of BCL2(Ala43Thr);(4)OE-MU:normal target cells,infec-ted with mutation type virus of BCL2(Ala43Thr).After Plasmid construction andtransfection for the PC3 cells,then followed by the wound healing assay.We measured the migration distance of each group at 0h , 6 h and 24 h and calculated the migration rate,using one-way ANOVA and LSD test to compare the differ-ence.Results:(1)No cell morphology difference was found among 4 groups.All cells grew well.(2)the cell clone formation rate determination results of control group was 30%,NC group was 20%,OE-wt group was 12% and OE-mu was 14%.(3)The migration rates of OE-wt group and OE-mu group were different with CON group at 6h (P =0.004,P =0.005 ).However,no difference between OE-wt group and OE-mu group at 6h(P>0.05).Conclusion:the polymorphism of BCL2 (Ala43Thr)OE-wt and OE-mu all had no influence on the migration of prostate cancer PC3 cell line.关键词
