摘要

Abstract

Objective:To explore the transfection and expression of green fluorescence protein reporter gene (GFP)in Human ligament fibroblasts mediated by lentivirus.And whether affect biophysical properties of ligament fibroblasts and provide theoretical basis for the next experiment.Methods:Human ligament fibro-blasts were Isolated and cultured by Tissue-adherent method and were transfected with lentiviral vector car-rying GFP.The GFP expression of ligament fibroblasts was observed under a fluorescence microscopy after 9 6 h.Growth curve was acquired by MTT and FACS to compare the difference of the growth rate between experimental and control groups.Results:Under the fluorescence microscope,green fluorescent was found partly after 9 6 h.And the GFP positive expression rate (transfection efficiency)was 9 5%.Flow cytometry testing results displayed that apoptosis of fibroblasts infected with GFP lentivirus were unaffected,as com-paredwith control MSC(P >0.05).The result of MTT measured growth curve showed that there fibro-blasts was no significant difference in the proliferative capability of chondrocytes between the experimental and control groups.Conclusion:The lentiviral vector containing green fluorescence protein reporter gene was infected Human ligament fibroblasts efficiently and expressed stably,which will provide a reliable tracer method,a basis for gene modification of ligament fibroblasts through the lentiviral vector.
