常压室温等离子体(ARTP)诱变及高通量筛选那西肽高产菌株
Mutation by using atmospheric and room temperature plasmas and high-throughput screening method for improving production of nosiheptide of Streptomyces actuosu
摘要
Abstract
A novel atmospheric and room temperature plasma (ARTP)mutagenesis system was used by combining with the high throughput screening of 48-well microplates and inhibition zone method to obtain high-yielding mutants. The results showed that the correlation coefficients between the ratio of inhibition zone diameter and mA408 ,and mA408 between fA408 reached 0. 534 and 0. 896,respectively. Through several rounds of ARTP mutation and high-throughput screening, three genetic stable mutants were obtained with increase by 50% relative contents of nosiheptide than that of the control strain finally. ARTP mutagenesis techniques as an effective way to obtain high-yield nosiheptide strains was compared with the traditional shake flask screening method,48-well microplate and inhibition zone method could significantly improve the screening efficiency of high nosiheptide-producing strains.关键词
