Abstract
Objective To investigate the effect and the possible mechanisms of progesterone on Aβ25-35-induced tau protein hyperphosphorylation in cortical neurons of primary rats. Methods Cortical neurons of SD rats were primarily cultured for 10 d, and then were divided into the control group, Aβ25-35 injury group and the progesterone protection group. The Aβ25-35 injury group was treated with 20 μmol/ L Aβ25-35 , while the progesterone protection group was treated with 20 μmol/ L Aβ25-35 and 1 μmol/ L progesterone. Western blotting was used to detect levels of tau phosphorylation and phosphorylation-AKt (p-AKt) expression in cortical neurons of rats. Results Phosphorylated levels on Ser404 and Thr231 points of tau protein in Aβ25-35 injury group were increased compared with those in control group, while the phos-phorylated levels of tau protein in progesterone protection group were decreased compared with those in Aβ25-35 injury group (P < 0. 05). The p-AKt expression was decreased in Aβ25-35 injury group compared with that in control group, while p-AKt expression was increased in progesterone protection group compared with that in Aβ25-35 injury group (P <0. 05). Conclusion Progesterone may attenuate Aβ25-35-induced tau protein hyperphosphorylation by increasing the AKt activity in cortical neurons of rats.关键词
