不同频率的周期性压力对兔软骨细胞影响的生物学研究
Effect and Mechanism of Cyclical Pressures of Different Frequencies on Rabbit Chondrocytes

Abstract
Objective:By observing cyclical pressures of different frequencies on rabbit chondrocyte proliferation and se -cretion of integrin α1β1, to study the effects and mechanism on rabbit chondrocytes .Method:Isolated fetal rabbit articular chondrocytes , the third-generation cells were seeded in plates .Set control group A ( normal culture ) and group B of different frequencies pressure (pressure 172kPa, frequency group B1 0.2Hz, group B2 0.5Hz, group B3 1Hz, group B4 2Hz, group B5 5Hz) .Pressure intervention of different frequencies 2 hours per day , 3 consecutive days after the intervention , to detect the proliferation of chondrocytes by Optical Density ( OD) , and ELISA assay was used to measure the integrin α1β1 level of culture supernatant.Result:OD value of group A was 1.08 ±0.13, group B1 was 1.19 ±0.11, group B2 was 1.23 ±0.08, group B3 was 1.35 ±0.06, group B4 was 1.45 ±0.09, group B5 was 1.37 ±0.12.Pressure groups compared with the control group was increased, the difference was statistically significant .There is significant difference between groups B , the peak appeared in B4 group.Group A of integrin α1β1 level was 3.02 ±0.26 pg/mL, group B1 was 3.28 ±0.19 pg/mL, group B2 was 3.43 ±0.11 pg/mL, group B3 was 3.77 ±0.24pg/mL, group B4 was 4.01 ±0.09 pg/mL, group B5 was 3.94 ±0.13 pg/mL.Integrinα1β1 value of pressure groups compared with the control group was increased , the difference was statistically significant ( P<0. 05) .There was significant difference between groups B; the peak appeared in group B 4.Conclusion: The same size ( 172 kPa) cyclic pressure stimuli on rabbit chondrocytes at different frequencies can promote chondrocyte proliferation and increase the integrins , but not the higher the frequency , the stronger the effect.In this study, the frequency of 2Hz has the strongest effect.关键词
