Effects of Serum Containing Bushen Zhuanggu Granules on Proliferation of Mesenchymal Stem Cells of Osteoporotic Rats
摘要
Abstract
Objective:To investigate the effect of serum containing Bushen Zhuanggu granules in-vitro proliferation of mesenchymal stem cells( MSCs)of osteoporotic( OP)rats. Methods:10 cases osteoporotic animal models were established in 6 months old SD rats by ovari-ectomy. MSCs were isolated from the OP rats and cultured in vitro. Cultivated MSCs were divided into 3 groups:OP group,OP+serum containing bushen zhuanggu granules group and OP+ blank serum group. The morphological difference of the MSCs in 3 groups was ob-served under inverted phase contrast microscope. Optical absorption value of MSCs was stained by methyl thiazolyl tetrazolium( MTT). The growth curve was measured. Result:In OP+serum containing bushen zhuanggu granules group,MSC was typical colony growth. The cell was long fusiform shape,which ecphyma of cell was slender and more. The cell proliferation was prosperity,population was increas-ing. But the rate of cell proliferation was to step down,when passage of cell was increasing. In OP group and OP+blank serum group, MSC body was comparatively large,meta-round shape. Ecphyma of cell shrink,the rate of proliferation was to step down,population was decreased. When passage of cell was increasing,step down of cell proliferation was significant. In OP + serum containing Bushen Zhuanggu granules group proliferative activity of MSCs was significantly promoted,the growth curve was significant difference in compar-ison with the control group. Conclusion:Serum containing Bushen Zhuanggu granules can enhance the proliferation of MSCs in OP rats. Reference citation:Tu Qiang,Cao Lu,Ding Huanwen,et al.
