摘要
Abstract
This paper introduced the working principle , conventional sample preparation and production requirements of laser scanning confocal microscopy , as well as the method of im-age acquisition , including the regulation of light path and light intensity adjustment , gain value and pinhole regulation , transmission light path and DIC settings .And introduced the confocal technique in biological research field , such as multi-track fluorescence collection , Z-stack scanning and three -dimensional construction , analysis of fluorescence colocalisa-tion and intensity , time sequence scanning and bleaching technology .This paper could effi-ciently provide technical reference for grasping the confocal operation skills .关键词
