高产淀粉酶黑曲霉菌种诱变选育及固态发酵条件研究
MUTATION BREEDING AND SOLID-STATE FERMENTATION CONDITIONS OPTIMIZATION OF ASPERGILLUS NIGER WITH HIGH YIELD OFΑ-AMYLASE

Abstract
The fungal amylase has the characteristic of mild hydrolusis conditions and acid-resistance, and a wide range of application. By composite mutagenesis of microwave mutagenesis and diethyl sulfate treatment, isolate with high yield ofα-amylase was screened and the solid-state fermentation conditions were optimized. The results showed that, A. niger MD-17 was obtained after 16 s microwave mutagenesis and 20 min diethyl sulfate treatment. The optimal solid-state fermentation conditions followed as bran:corn flour=7:3;(NH4)2SO4:0.6%;initial pH 5;solid material:water 1:1.5. Fungalα-amylase enzyme activity could reached the highest 6011 U/g.关键词
