Abstract
Objective To explone the expression and significance of Sox2 and Oct4 in human glioma tissue of different pathological grade. Methods Immunohistochemical technique and tissue microarray were used to detect the expression of Sox2 and Oct4 in 45 cases of human glioma tissue and 5 cases of normal brain tissue. Results The expression rate of Sox2 was (2.61±1.33)%in normal human brain tissue,the results were negative,and the positive expression rate of Sox2 in human glioma tissue of WHO Grade Ⅰ,II,Ⅲ,Ⅳ was (3.95%±3.40)%,(18.22%±5.16)%,(20.74%±5.14)%,(60.15%±7.82)% respectively,and there was a statistical difference of the expression of Sox2 in normal human brain tissue and human glioma tissue (F=87.9,P<0.01),there was a statistical difference of betweent the gradeⅠ,II and gradeⅢ,Ⅳ (P<0.01).Oct4 in normal human brain tissue had no expression,the positive expression rate of Oct4 in human glioma tissue of WHO Grade Ⅰ,II,Ⅲ,Ⅳwas (1.25±3.82)%,(37.20±9.12)%,(51.39±9.72)%,(63.15±9.05)%respectively,and there was a statistical difference of the expression of Oct4 in normal human brain tissue and human glioma tissue (F=67.7,P<0.01), there was a statistical difference of betweent the gradeⅠ, IIand gradeⅢ, Ⅳ (P<0.01).The expression intensity of Sox2 and Oct4 were increased with the pathological grade elevating,while in normal human brain tissue showed no expres-sion;Sox2 expression in human glioma tissue was positively correlated with that of Oct4 (r=0.619,P<0.01).There was no statistical difference of the positive expression rate of different sex and age in human glioma tissue (P<0.05). Conclusion The expression of Sox2 and Oct4 in human glioma tissue is positively related with the malignant degree of the tumor and it is suggested that Sox2 and Oct4 may play an important role in the occurence and development of the human glioma tis-sues and its high expressions in human glioma tissue may be one of the reasons of higher degree of maglignancy and in-vasive growth in human glioma.关键词
