桦褐孔菌漆酶基因片段的克隆与序列分析∗
Cloning and Sequence Analysis of Laccase Gene Fragments from Strains of Inonotus obliquus
摘要
Abstract
Laccase gene fragments of 11 strains of Inonotus obliquus and several other species in the same genus were cloned by using degenerate primers, and its structure and homology were also ana⁃lyzed. The results indicated that two introns, the positions of which were highly conserved, were contained in all 11 PCR-amplified products, and conserved copper⁃binding sites ( His) were shown in the alignment of amino acid sequences. High similarities were reveled in both DNA and amino acid sequences, and the NJ trees constructed on the basis of the two sequences displayed that all I. obliquus strains expect Chaga were grouped together, suggesting this laccase gene fragment could be applied in species identification of I. obliquus.关键词
