Abstract
Objective To explore the molecular mechanism of IL-1 7-induced the secretion of IL-10 by mono-cytes.Methods The peripheral blood was collected from the healthy individuals undergoing the health physical ex-amination in our hospital physical examination center,the mononuclear cells were isolated by ficoll density gradient centrifugation,and the CD14 positive monocytes were purified from mononuclear cells by using immunomagnetic beads.In vitro CD14 positive monocytes were stimulated by IL-1 7,or signal pathway inhibitors were pre-incubated for 1 h and then stimulated by IL-1 7,the level of IL-10 was detected by enzyme-linked immunosorbent assay.Results after IL-1 7 stimulation,the secretion of IL-10 by CD14 positive monocytes was significantly increased (P <0.01), which is in a dose and time dependent manner.However,IL-1 7-induced the secretion of IL-10 was significantly de-creased while adding the NK-κB inhibitor (P <0.05),but the MEK1/2 inhibitor,p38/MAPK inhibitor and JNK in-hibitor had no obvious inhibiting effect.Conclusion IL-1 7 could induce monocytes to secret IL-10 by the NK-κB sig-nal pathway and thus plays the immunosuppressive effect.关键词
