Abstract
Objective:To probe the biological properties of the constructed rabbit bone marrow mesenchymal stem cell sheet and its advantages for tissue engineering. Method:BMMSCs were obtained from Japanese white rabbits and used to construst BMMSCs sheet with simple methods. Inverted microscope and HE staining were employed for morphological observation.MTT assay and osteogenic/adipogenic induction were examined for the proliferation and differentiation properties of the cell sheet. Related genes'mRNA expression was examined by RT-PCR. Result:After 10~14 days in vitro culture of rBMMSCs,a white membranaceous sheet was formed and lifted by aseptic forceps .rBMMSCs grew in multilayer and retained their spindle shape observed under microscope. HE staining showed abundant extracellular matrix (ECM).The cell density increased in the culture. The results of cell sheet's osteogenic/adipogenic induction were significantly better than rBMMSCs. Compared with rBMMSCs,gene expression of ColⅠand FN were significantly up-regulated in cell sheet,which was indicat-ed by RT-PCR. Conclustion:Compared with rBMMSCs in common culture,cell sheet had more advantages for tissue engi-neering.关键词
