Effect of celecoxib, a cyclooxygenase-2 inhibitor, on the expression of erythropoietin in retina of diabetic rats

Abstract
Objective To observe the effect of cyclooxygenase-2 inhibitor on the expression of erythropoietin ( EPO) in diabetic rats retina and the effect of celecoxib on prevention of early diabetic retinopathy .Methods Forty SD male rats were randomly divided into four groups:the normal group , diabetes group , control group and experimental group , 10 in each group.Diabetes model was established by intraperitoneal injection of streptozotocin (STZ) (50 mg/kg).All of the rats were sacrificed at 3 months.Retinae were HE stained to observe morphological changes , and SYBR real-time fluo-rescent quantitative PCR was used to detect COX-2 and EPO mRNA expression changes .Results After 3 months, HE staining showed structural changes in diabetes group and control group .There was no difference in COX-2 or EPO mRNA expression between diabetes group and the control group ( P <0.05), but between diabetes group and other two groups ( P <0.05).Conclusions Celecoxib could effectively promote the expression of EPO , thus can be used for early preven-tion and treatment for diabetic retinopathy .关键词
