Relationship between the altered expression of oxidative damage repair gene HOGG1 and age-related cataract

Abstract
Objective To observe the altered expression of HOGG 1, a base excision repair enzyme for repairing oxidative DNA damage in anterior lens capsule epithelial cells (LECs) in age-related cataract (ARC) patients, and to ex-plore potential biological roles of HOGG 1 in the pathological mechanisms of ARC .Methods LECs samples of cortical cat-aract, nuclear cataract and posterior cataract were collected .Clear lens samples were collected as control .We conducted RT-PCR and immunochemistry to measure the expression of HOGG 1.Results We found that the expression of HOGG 1 was significantly higher in LECs of ARC patients than in control LECs ( F =107.62, P <0.01), but no differences be-tween cortical , nuclear or posterior subcapsular cataracts .Immunofluorescence showed that positive staining of HOGG 1 were seen in cell nucleus and cytoplasm in either group , but was stronger in cataractous LECs .Conclusion The mRNA and protein expression of HOGG 1 increased in ARC LECs , which may provide theoretical basis for early diagnosis and treatment of ARC.关键词
