针刺介入时机对脑梗死大鼠能量代谢和脑源性神经生长因子、酪氨酸激酶受体B的影响
Effect of acupuncture intervention time on energy metabolism and BDNF,TrKB in rats with cerebral infarc-tion
摘要
Abstract
Objective To research the effects of acupuncture intervention time on energy metabolism and the expression of BDNF and TrKB in rats after ischemic stroke and explore the mechanism of acupuncture treatment and intervention time .Methods The method of permanent focal cerebral ischemia of middle cerebral artery was estab -lished in rats.Two hundreds of model were selected and divided into 5 groups:sham-operation group,model-group,2 h acupuncture group,3 d acupuncture group,7 d acupuncture group.The sodium,potassium-adenosine triphosphatase (Na+,K+ATPase),calcium and magnesium-adenosine triphosphatase (Ca2+,Mg2+ATPase) and the number of BDNF,TrKB positive nerve cells were measured at 1 d,3 d,7 d and 14 d of postoperation .Results The Na+,K+-ATPase,Ca2+,Mg2+-ATPase activity in brain tissue of rats were sharply decreased after ischemia ,lowest at 1d,along with the changes in the extension first and then drop time .ATPase activity in acupuncture group increased significantly in 1,3,7,14 d than those in the model group (P<0.01,P<0.01).2 h group was higher than 3 d and 7 d groups (P<0.01,P<0.05),the difference was statistically significant.The BDNF,TrKB positive nerve cells were gradually increased with time(P<0.05).The BDNF,TrKB positive neural cells of each acupuncture group were increased gradually with time(P<0.01,P<0.05).The number of TrKB positive cells in 2h acupuncture group was higher than that in 3 d,7 d acupuncture group(P<0.01,P<0.05).Conclusion Acupuncture can improve energy metabolism of ischemic brain tissue ,increase the positive neurons cell number of BDNF and TrKB ,repair and protecte the brain injury of ischemic ,the better therapeutic effect the earlier acupuncture treatment .关键词
