Abstract
Objective:To study on CCK-8 method for detection of Achyranthes bidentata alcohol induced by extracts of rabbit bone marrow mesenchymal stem cells ( BMSCs) optimal detection conditions.Methods: With rabbit BMSCs isolation, culture and amplifica-tion, the amplification of the subculture, and take the fourth generation cells used in the experiments.Comparison of CCK-8 absorption spectrophotometry method of difference value in different incubation time and different wavelength detection conditions, detection of Achy-ranthes bidentata alcohol research method of CCK-8 induced by extracts of rabbit BMSCs optimal testing time and the best detection wavelength.Results:Achyranthes bidentata alcohol extract could obviously induce rabbit bone marrow mesenchymal stem cells prolifera-tion induced by BMSCs, the feasibility of rabbit cartilage differentiation had strong.CCK-8 method for detection of Achyranthes bidentata alcohol induced by extracts of rabbit BMSCs optimal testing time to join the CCK-8 reagent after incubation of the 2h, the best detection wavelength of 450nm.Suitable for detection of cell number range of3 ×103/ml~4 ×10^4/ml.Conclusion:CCK-8 method in after 2h incubation and enzyme mark instrument 450nm wavelength detection of Achyranthes bidentata alcohol extract induced rabbit BMSCs high-est sensitivity.关键词
