Abstract
Objective To investigate the effect of salidroside on the proliferation and apoptosis of human melanoma cell line A375.Methods The A375 cells were cultured with different concentrations of salidroside (0,0.01,0.1,1,10, and 100 μg/mL) .The inhibition of cellular proliferation was determined by MTT .The apoptosis was assessed by the An-nexin-V-FIFT/PI affinity assay.The mRNA levels of CDk4, CDk2, cyclinE and cyclinD1 were detected by RT-PCR.Re-sults Compared with the negative control group , the proliferation inhibition rate of A 375 cells of the salidroside group was significantly higher(P <0.05).The cells of the salidroside group were mainly at the G0/G1 phase, and the cells in-creased in the S phase and decreased in the G 2/M phase (P<0.05).The mRNA expression of CDk2, CDk4, cyclinD1 and cyclinE in the salidroside group was inhibited , which indicated that salidroside limited the proliferation of A 375 cells by regulating the cell cycle, and the difference was statistically significant (P<0.05).Conclusion Salidroside has the effect of inhibiting the proliferation and enhancing the apoptosis of human melanoma cell line A 375 .关键词
