阿霉素对大肠癌 SW480细胞周期及周期相关蛋白表达的影响
Effect of adriamycin on cell cycle and cycle-related protein expression of colorectal cancer SW480 cells

Abstract
Objective To investigate the effect of adriamycin on the cell cycle and cycle-related protein expression of colorectal cancer SW480 cells.Methods SW80 cells cultured in vitro were randomly divided into groups A (blank control group), B, C, D and E, which were treated with different concentrations of adriamycin (0μg/mL, 2.5μg/mL, 5.0μg/mL, 10 μg/mL and 20 μg/mL).After being cultured for 12, 24, 48 and 72 h, the growth inhibition rate of each group was detected by using methyl thiazolyl tetrazolium (MTT) method.After being cultured for 48 h, the cell cycle of SW480 cells in each group was analyzed by flow cytometry , and the expression of cycle-related proteins (cyclin B1, cyclin A) of each group was detected by Western blotting .Results The inhibitory rate of SW480 cell growth affected by adriamycin was in a dose-and concentration-dependant manner .After being treated by different concentrations of adriamycin for 48 h, the percentage of cells in the G 0/G1 phase of the groups D and E was lower , the percentage and apoptosis index in the SubG1 phrase of groups D and E were higher than those of groups A , B and C (all P<0.05).The expression level of cy-clinBl in the group B and group C was higher than that of group A , but the expression level of cyclinBl in the group D and group E was lower than that of group A (all P<0.05).The expression of CyclinA gradually decreased with the increasing concentration of adriamycin (P<0.05).Conclusion Adriamycin can block the cell cycle at the G 0/G1 phase of SW480 cells, and effect the expression of cyclin B 1 and cyclin A .关键词
