结肠癌组织中Ku70、转录因子Sp1的表达变化及意义
Expression of Ku70 and transcription factor Sp1 in colorectal cancer and its significance

Abstract
Objective To detect the expression of Ku70 and specificity protein 1 ( Sp1 ) in colorectal cancer tissue samples.Methods Immunohistochemistry and reverse transcription-polymerase chain reaction( RT-PCR) were used to de-tect the proteins and mRNA expression of the Ku70 and Sp1 in colorectal cancer samples and matched tumor-adjacent tissue samples respectively.The correlation beween the expression of Ku70 and Sp1 and clinical pathological parameter was also analyzed.Results The positive expression rate of Ku70 in tumor tissues and matched tumor-adjacent tissues was 68.9%(42/61) and 36.1%(22/61) respectively, the difference was statistically significant(P<0.05).The positive expression rate of Sp1 was 78.6%(48/61) and 26.2%(16/61) respectively, the difference was statistically significant(P<0.05). The relative expression of Ku70 mRNA in tumor tissues and matched tumor-adjacent tissues was 1.184 ±0.410 and 0.845 ±0.424 respectively, the difference was statistically significant(P<0.05).The relative expression of Sp1 mRNA was 1.280 ±0.311 and 0.912 ±0.362, the difference was statistically significant(P<0.05).The correlation of Ku70 mRNA and Sp1 mRNA was positive(r=0.398, P=0.029).The expression levels of Ku70 and Sp1 were associated with tumor size, tumor differentiation, lymph node metastasis, TMN stage(all P<0.05).Conclusions The expressions of Ku70 and Sp1 in colorectal cancer are both higher than those in tumor-adjacent tissue, and are highly associated with the metastasis and infiltration in colorectal cancer.They may become new biomarkers in the occurrence and development of colon cancer .关键词
