法舒地尔预处理对大鼠缺血再灌注损伤肝脏的保护作用及机制探讨
The protective effect and mechanism of Fasudil preconditioning on hepatic ischemia-reperfusion injury in rats

Abstract
Objective To investigate the protective effect and mechanism of Fasudil preconditioning on hepaitc ische -mia-reperfusion injury in rats .Methods 30 adult SD rats were randomly divided into sham group ( S group ) , ischemia-reperfusion group ( IR group:produced by total inflow occlusion for 30 minutes) , fasudil group ( FAS group:fasudil was in-traperitoneal injected 30 minutes before ischemia ) , 10 rats in each group .The hepatic artery was completely block with no damage clamp for 30 minutes in IR group and FAS group, and the clamp was removed 2 hours later.In FAS group, fasudil was intraperitoneal injected with the dose of 10 mg/kg before the artery was blocked , and in IR group , saline was intraper-itoneal injected with the same dose .In S group, we just dissected the hepatic ligaments and exposed hepatic hilar vessels without blocking blood flow .After the models had been established , serum ALT, AST and the liver tissue superoxide dis-mutase (SOD) activity, malondialdehyde (MDA) content and myeloperoxidase(MPO) activity were measured after reper-fusion for 2 hours.And the liver pathological changes were observed .Results Compared with the S group , the serum ALT, AST levels in IR group and FAS group were significantly higher (all P<0.05).Compared with IR group, the serum ALT, AST levels in FAS group were lower (all P<0.05).Compared with the S group, IR group and FAS group SOD ac-tivity decreased (all P<0.05), MDA content and MPO activity increased (all P<0.05).Compared with IR group, FAS group SOD activity increased (P<0.05), MDA content and MPO activity decreased (all P<0.05).For the pathological changes of the liver , the hepatic cords distributed orderly , and the liver cell morphology was normal in S group .After is-chemia-reperfusion for 2 h, the hepatic cell cord became disorder , and the liver cells were swelling , with vacuolar degener-ation, and the alignment of sinusoidal space narrowed or disappeared , the inflammatory cells were infiltrating in IR group . Compared with IR group ,the disorderness of hepatic cords , swelling of liver cells were reduced in FAS group , and the infil-tration of inflammatory cell were less in FAS group .Conclusion Effect of Fasudil preconditioning on ischemia-reperfusion injury of the liver is protective and its mechanism may involve the improvement of the body 's resistance to oxidative stress and reduce neutrophilic infiltration in liver tissues .关键词
