不同心功能糖尿病心肌病大鼠瞬时外向钾电流的变化及意义
Significance of myocardial Ito changes in different cardiac functions of rats with diabetic cardiomyopathy

Abstract
Objective To investigate the variations and significance of the transient outward potassium current ( Ito) in different cardiac functions of rats with diabetic cardiomyopathy (DCM).Methods We established the DCM animal models by intraperitoneal injection of STZ in rats (DCM group), and heart failure models by ligation of the left coronary ar-tery in DCM rats ( DCM with heart failure group ) .Rats in the control group were treated with sham-operation .Whole-cell patch clamp technique was used to record and compare the unicellular changes of Ito current in the left ventricular myocytes of the three groups , respectively .Results The left ventricular dilation and heart dysfunction were only found in the DCM with heart failure group after ligature of the coronary artery in 3th week.Significant difference was found between the DCM with heart failure group and the DCM group, control group (all P<0.05).Compared with control group, Ito density of the left ventricular myocytes was lower and it happened early in the DCM with heart failure group [(12.05 ±9.02) pA/pF vs (32.11 ±4.52) pA/pF, at +60 mV, P<0.05;(7.70 ±9.21) pA/pF vs (38.65 ±5.05) pA/pF, at +70 mV, P<0.05].The Ito density was lower in the DCM group as compared with that of the control group [(15.22 ±9.13) pA/pF vs (38.65 ±5.05) pA/pF, at +70 mV, P<0.05].The more depressed I-V curves of Ito were found in both DCM and DCM heart failure groups as compared with the control group , but the changes between DCM and DCM heart failure group were not statistically significant (all P>0.05).Conclusion As the cardiac function decreases , the Ito current density of DCM rats decreases , which may be one of the important ion bases for the occurrence of arrhythmia in DCM patients .关键词
