ADAM17对前列腺癌细胞 TGF-α、EGFR 的表达影响
Effects of ADAM17 on TGF-αand EGFR in prostate cancer cells

Abstract
Objective To investigate the relationship between a disintegrin and metalloprotease 17 (ADAM17) and transfor-ming growth factor-α(TGF-α) activation in prostate cancer cells DU145.Methods We cultured the prostate cancer DU145 cells. Then, the cells were divided into the control group, NC group, ADAM17-siRNA1 group and the combination group.ADAM17 siRNA was transfected into both the ADAM17-siRNA1 group and combination group.The NC group was transfected with non-DAM17 related sequence siRNA.After the combination group was transfected for 72 h, 20 g/L TGF-αwas added.After 72-hour transfection, the cells of the control group, NC group and ADAM17-siRNA1 group were collected.Western blotting was used to detect the protein ex-pression of ADAM17, epidermal growth factor receptor (EGFR) and pEGFR.ELISA was used to detect the level of TGF-αprotein. We collected the supernatant intervened for15 mins of TGF-αfrom the control group, ADAM17-siRNA1 group and the combination group, and Western blotting was used to detect the protein expression of EGFR and pEGFR.Results The protein expression of AD-AM17, pEGFR and TGF-αin the ADAM17-siRNA1 group was lower than that of the control group (all P<0.05).There was no sig-nificant difference in EGFR expression between the control group, NC group and ADAM17-siRNA1 group (P>0.05).The expression of pEGFR in the combination group was higher than that in the control group and the ADAM17-siRNA1 group (P<0.05).There was no significant difference in the EGFR expression between the control group, ADAM17-siRNA1 group and the combination group (P>0.05).Conclusion The expression of ADAM17 in prostate cancer DU145 cells is correlated with the activation of TGF-αand EG-FR, and ADAM17 activates pEGFR expression by activating TGF-α.关键词
