Medium optimization for α-pinene production with response surface analysis

Abstract
Response surface analysis was used to optimize for α⁃pinene production. Initially, MD beef extract powder was determined to be the best nitrogen source for α⁃pinene production, then Plackett⁃Burman ( P⁃B ) experiment was used to evaluate the eight nutritional variables to screen medium component that significantly influence the α⁃pinene production and their optimal levels. Results showed that beef extract powder,pH and citric acid were significant factors. Then the path of steepest ascent was used to approach the optimal region of the above three factors. In the third step,the concentration of beef extract powder,pH and citric acid was further optimized using central composite designs and response surface analysis. As the result,the concentration ofα⁃pinene increased from 1�710 to 7�053 mg/L. With the optimized medium,the yield of α⁃pinene increased about 3⁃fold.关键词
