D-阿拉伯糖醇高产酵母菌株的快速筛选方法
A rapid screening method of yeast strains for hight yield production of D-arabitol

Abstract
A high throughput screening method that using high sugar medium for preliminary screening and paper chromatography in combination with high performance liquid chroma‐tography for further screening was established to selectively breed the high‐yield D‐arabitol strain by low‐energy N+ ion implantation .The expand system ,sample volume and chromoge‐nic method of paper chromatography were optimized .The results indicate that optimal condi‐tions are as follows :eluent∶ n‐butanol∶pyridine∶water=14∶3∶3 ,sample volume:1 .5μL , chromogenic agents 50% acetone solution of silver nitrate and 50% sodium hydroxide solu‐tion in ethanol .High‐yield D‐arabitol producing strains were obtained based on the size of significant colored spots ,then high performance liquid chromatography was used to verify the result .The results show that this method can be used to screen quickly D‐arabitol high‐yield strains from large quantities of mutated strains .The high‐yield strain has good genetic stabil‐ity and its production was 88 .23 g/L .关键词
