慢病毒介导干扰 TM4SF1基因表达对血管内皮细胞体外生物学行为的影响
Effect on biological behavior in vitro of human umbilical vein endothelial cells by down﹣regulation of gene TM4SF1 expression with RNAi
摘要
Abstract
Objective:To investigate the effect of down﹣regulatedgene TM4SF1 expres﹣sion by RNAi on proliferation,cell cycling,migration and angiogenesis of human umbilical vein endothelial cells ( HUVEC ). Methods: Lentiviral vector to silence gene TM4SF1 ( LV﹣TM4SF1﹣RNAi) and itscorresponding control (LV﹣CON﹣RNAi) were constructed to infect HU﹣VECs. Flowcytometry sorting was applied to harvest HUVEC infected with LV﹣TM4SF1﹣RNAi or LV﹣CON﹣RNAi,which were then compared with blank HUVECs on capabilities of proliferation, migration and angiogenesis. Results:Lentivirus infected HUVEC efficiently and stably,com﹣pared with the LV﹣CON﹣RNAi and HUVEC group. The expression of TM4SF1 mRNA and pro﹣tein in LV﹣TM4SF1﹣RNAi was significantly lower than those in LV﹣CON﹣RNAi and HUVEC group(P<0. 05). The proliferation of LV﹣TM4SF1﹣RNAi group(0. 59 ± 0. 06) after inoculated 72h was significantly lower than LV﹣CON﹣RNAi and HUVEC group (0. 94±0. 04,1. 04±0. 05)(P<0.05).TheproportionofG0/G1phaseinLV﹣TM4SF1﹣RNAigroup[(69.30±5.48)%]wassignificantlymorethanthoseofLV﹣CON﹣RNAiandHUVECgroup[(54.40±4.09)% and(51.69±4.16)%](P<0.05).ThemigrationofLV﹣CON﹣RNAigroup(14.20±4.59) was (si7g4ni.f4ic0a±nt1l6y.l0o6wer than those of LV﹣CON﹣RNAi and HUVEC group[(70. 60 ±17. 75) and)](P<0. 05). The angiogenesis of LV﹣CON﹣RNAi group (3. 33±1. 52) was s2i.g0n8ificantly lower than those of LV﹣CON﹣RNAi and HUVEC group (19. 66±1. 52 and 22. 66±RNA)(P<0. 05). There was no statistically difference between HUVEC group and LV﹣CON﹣tion aingdroaunpg.iConclusion:Down regulation of gene TM4SF1 can inhibit the proliferation,migra﹣ogenesis of HUVEC,suggesting that gene TM4SF1 is a potentialtarget for anti﹣an﹣giogenic therapy.关键词
