Abstract
Objective:To explore the effects of evodiamine on the proliferation of ovari-an cancer SKOV3 cells and the underlying mechanisms. Methods:MTT assay was used to in-vestigate the cytotoxic effects of evodiamine on SKOV3 cells. Annexin V-FITC/PI staining and Western blot were used to investigate the evodiamine-induced apoptosis in SKOV3 cells. A tumor xenograft model was used to investigate the inhibitory effects of evodiamine on SKOV3 cells in vivo. Results:Evodiamine significantly inhibited the proliferation of SKOV3 cells in vitro,in a time-and dose-dependent manner. Evodiamine significantly induced apoptosis in SK-OV3 cells,accompanied by an increase of Annexin V-positive cells,as well as a evidently cleav-age of caspase-3 and PARP. Daily treatment of evodiamine led to a substantial inhibition of tumor growth in ovarian cancer xenograft model with concomitant cleavage of caspase-3 and PARP. Conclusion:The in vitro and in vivo results demonstrate that evodiamine efficiently sup-presses the proliferation of ovarian cancer SKOV3 cells and suggest that evodiamine may be a potential therapeutic agent for the treatment of ovarian cancer.关键词
