石榴皮醇提物对胃癌SCG-7901细胞增殖的影响
Effect of pomegranate peel granatoline extracts on proliferation of gastric cancer SGC-7901 cells
摘要
Abstract
Objective To approach the effect of pomegranate peel granatoline extracts on proliferation of gastric cancer SGC-7901 cells and its anti-cancer mechanisms. Methods The gastric cancer SCG-7901 cells were cultured in vitro and the pomegranate peel granatoline was prepared whose final concentration was 100 ,80,60,40,20,10 μg/mL respectively. Another negative control group was set up without given medicine (0μg/mL). It adopted MTT and cloning assays to observe and compare the effect of pomegranate peel granatoline of on proliferation and cloning capabilities of gastric cancer SGC-7901 cells. The mor-phologic changes of SCG-7901 cells acted by extracts for 48 hours were observed by inverted phase contrast microscope. The peri-odic variation of SCG-7901 cells treated with extracts for 48 hours were analyzed by flow cytometry(FCM). Results The pomegranate peel granatoline extracts displayed obvious inhibition effect on SCG-7901 cells,being distinctive concentration-dependent and time-dependent,and it significantly inhibited the formation rate of cells colony. Acted by different-concentration extract solution, the SCG-7901 cell numbers decreased with death of partial cells and karyopy knosis as well as the cell cycle changed. Compared with the negative control group,the ratio of S-phase cells was decreased and G0/G1-phase increased,as well as an obvious block in G0/G1 phase. Conclusion The pomegranate peel granatoline extracts may inhibit the proliferation of gastric cancer SGC-7901 cells,preventing the cells from G1 phase to S phase.关键词
