PPARγ、PTEN、Akt在胃癌中的表达及其与胃癌生物学行为的关系
Relationship between expression of PPARγ,PTEN and Akt and biologic behaviors of gastric carcinoma
摘要
Abstract
Objective To investigate the relationship between expression of peroxisome proliferator-activated receptors γ(PPARγ),phosphatase and tensin homology deleted on chromosome 10 (PTEN)and Akt and biological behaviors of gastric carcinoma. Methods The expression of PPARγ,PTEN and Akt mR-NA in 60 cases of gastric carcinoma and the corresponding non-carcinoma tissues and 20 cases of normal gastric tissues were detected by reverse transcription polymerase chain reaction (RT-PCR ). The correlation of PPARγ,PTEN and Akt expression with tumor differentiation grade,size,lymph node metastasis,distant me-tastasis,depth of invasion,TNM stage and patients’gender and age were analyzed. Results PPARγand Akt expression in gastric carcinoma were significantly higher but PTEN expression significantly lower than in corre-sponding non-carcinoma tissues and normal gastric tissues (P<0.05 ). There was no significant difference in the expression of PPARγ,PTEN and Akt mRNA in non-carcinoma tissues and normal gastric tissues. PPARγ, PTEN and Akt expression in gastric carcinoma were significantly associated with TNM stage,serosal invasion, lymph node metastasis and distant metastasis (P<0.05 ),but not correlated with the gender,age,tumor size and differentiation grade. The expression of PPARγwas negatively correlated with PTEN (rs =-0.492,P<0.05)but positively with Akt (rs =0.623,P<0.05). Moreover,the expression of PTEN was negatively cor-related with Akt (rs =-0.565,P<0.05). Conclusion PPARγand Akt are highly expressed in gastric car-cinoma,while PTEN is lowly expressed in gastric carcinoma and there is a significant correlation between them.They may serve as markers in the diagnosis of gastric cancer and estimation of prognosis.关键词
