Abstract
Objective To analyze the activity and receptor expression of peripheral natural killer (NK)cells in children with β-thalassemia major and investigate underlying mechanism of the transfusion-in-duced immunodepression. Methods Peripheral blood samples were obtained from 20 children withβ-thalasse-mia major,20 β-thalassemia minor and 20 healthy volunteers. Expression levels of CD107a,activating recep-tors (NKP30,NKP46 and NKG2D)and inhibitory receptors (NKG2A and CD158a)within circulating NK cells were detected by multicolor flow cytometry. The level of IFN-γsecreted by NK cells was measured by in-tracellular cytokine staining. Results Compared with β-thalassemia minor and healthy counterparts (all P<0.05),the cytotoxicity (CD107a expression)of circulating NK cells was remarkably down-regulated whereas NKG2A expression,as an inhibitory receptor of circulating NK cells,was significantly up-regulated in children with β-thalassemia major (all P<0.05 ). However,no statistical significance was observed in the expression of NKP30,NKP46,NKG2D,CD158a and IFN-γ(all P>0.05)among three groups. Conclusion NKG2A, as an inhibitory receptor of NK cells,may play a key role in the transfusion-induced immunodepression of NK cells in children with β-thalassemia major.关键词
