Abstract
Objective To investigate the effect of atorvastatin on pathology and expression of serum tumor necrosis factor ( TNF-α) and high mobility group protein 1 ( HMGB-1 ) in lung tissue of rats with sepsis .Methods Seventy-five healthy SD rats were randomly divided into sham operation group , sepsis group and intervention group with 25 rats in each group.In each group, after 12, 24, 36, 48, 72 h, 5 rats from each group were sacrificed and blood of the heart and lung tis-sue were sampled , lung tissue routine HE staining were perfomred to observe the pathological changes , serum TNF-αlevel were determined with ELISA method , expression of HMGB-1 were detected by Western Blot method .Results A few inflam-matory cells were found in the sham operation group;Lung tissue and alveolar edema , infiltration of inflammatory cells and red blood cells were found in sepsis group;intervention group ’ s inflammatory cells in lung tissue were less than sepsis group , the alveolar structure is complete , the pathological changes were obviously relieved .Sepsis group and intervention group ’ s lung tissue’s W/D were higher than those in the sham operation group ( P <0.01), and they were decreased significantly after the 36 h in the intervention group, and lower than sepsis group ( P <0.05).In each time point, TNF-αand HMGB-1 ex-pression of intervention group and sepsis group were significantly higher than those in the sham operation group ( P <0.05), and the expression levels were significantly lower in intervention group than in the sepsis group ( P <0.05), After 36 h, the expression level of TNF-αand HMGB-1 in the intervention group and sepsis group were decreased ( P <0.05).Conclusion Atorvastatin can improve the pathological inflammatory lesion ,reduce the serum TNF-αand HMGB-1 expression of lung tis-sue in rats with sepsis .关键词
