Abstract
Objective To investigate the effect of hesperetin in anti apoE-/-mice atherosclerosis and its mechanism. Methods Forty eight-week-old apoE- / -mice were randomly divided into four groups, normal diet group ( NC) , atheroscle-rosis high-fat diet group (WD), hesperetin high dose (100 mg/kg) group (H-HES), hesperetin low dose (50 mg/kg) group ( L-HES) , each group with 10 mice, by using of orally administered hesperetin, according to each mouse body weight to give different dose, the mice were sacrificed after 12 weeks and orbital venous blood were sampled, automatic biochemical analyzer was used to detect the serum triglyceride, total cholesterol, low-density lipoprotein cholesterol and high-density lipoprotein cholesterol levels, and using ELISA kit to detect the serum tumor necrosis factor ( TNF-α) and monocyte chemoattractant pro-tein-1 (MCP-1)'s concentration in these apoE-/-mice. After anesthetized, mice'left ventricle were perfused with paraformal-dehyde, aortic tree oil red O staining were used to observe the percentage of plaque, while the aortic sinus were sampled and by using immunofluorescence to observe the infiltration of macrophages in the plaque. Results Compared with NC, WD group, H-HES and L-HES group's serum TC, TG, LDL-C concentrations were significantly increased, HDL-C significantly de-creased ( P <0. 05), H-HES and L-HES group's TC, TG, LDL-C level was lower than that in WD group ( P <0. 05). High fat and high cholesterol diet for 12 weeks, the lesions of aortic atherosclerosis in group WD was very significant,AS plaque area percentage of the aorta was (0. 143 0 ± 0. 060 4);and the area percentage in H-HES group and L-HES group were (0. 040 4 ± 0. 017 5) and (0. 025 2 ± 0. 019 5), compared with the WD group, H-HES group and L-HES group's aortic AS were signifi-cantly decreased, the difference was statistically significant ( P <0. 05). Macrophages in atherosclerotic lesion of aortic si-nus in group WD was increased, after 12 weeks of hesperetin treatment, the H-HES and L-HES group's aortic sinusplaque's CD68 macrophage infiltration were decreased ( P < 0. 05). WD group 's serum MCP-1 and TNF-α concentration were (807. 33 ± 122. 46) and (9. 87 ± 1. 75) μg/L, which were higher than H-HES and L-HES group's (365. 83 ± 66. 31) and (6. 34 ± 0. 65) μg/L and (405. 74 ± 46. 21), (9. 48 ± 0. 82) μg/L ( P <0. 05). Conclusion Hesperetin has significant effect on decreasing the blood lipid, also has anti atherosclerosis effect, mainly through its anti-inflammatory pharmacological effects.关键词
