Abstract

Objective: To study the role of MEK-ERK signaling pathway in ATRA-induced differentiation of APL cells and its potential mechanisms. Methods: APL cell line NB4 was used as in vitro model. The cell differentiation was evaluated by cell surface differentiation antigen CD11b, nitroblue-tetyrazolium (NBT) reduction assay and cellular mor-phology. The activation of MEK and ERK as well as the protein expression levels of PU.1, C/EBPβ, C/EBPε and PML-RARα were assayed by Western blotting. Results: MEK-ERK signaling pathway was activated at the early period of ATRA treatment and the activation persisted for 48 h. Attenuation of activation of MEK resulted in declination of percent-age of ATRA-induced CD11b positive cells from (87.50±4.16)%to (38.01±2.79)%. Meanwhile, A540 value of NBT reduc-tion assay was also decreased from 0.507±0.009 to 0.250±0.010. Both were statistically significant (P<0.01 and P<0.001 respectively). Furthermore, most of cells restored to characteristic morphology of APL cells. The increased protein levels of PU.1, C/EBPβ and C/EBPε induced by ATRA treatment were suppressed. Conclusions: ATRA could induce differentia-tion of APL cells through regulation of protein level of PU.1, C/EBPβand C/EBPεby MEK-ERK signaling pathway.
