Effect of homocysteine on proliferation and production of collagens in rat vascular smooth muscle cells

Abstract
Objective: To investigate the effect of different concentrations of homocysteine (Hcy)on proliferation of rat vascular smooth cells (vSMCs) and correlations with production of collagen types Ⅰ, Ⅲ, Ⅳ and Ⅵ. Methods: Rat vascular smooth muscle cells (vSMCs) were treated with different concentrations of Hcy, and at different timepoints the cell viability was evaluated by MTT, cell proliferation by BrdU and flow cytometry, and collagens were measured by ELISA. Results: vSMCs proliferation increased significantly at concentrations of Hcy 0.025-0.500 mmol/L after 12, 48 h of treatment. Production of collagen types Ⅰ, Ⅳ were increased with the increase of concentrations of Hcy in a dose-de-pendent manner, while type Ⅲ only increased slightly without statistical significance. Type Ⅵ collagen was significantly reduced at high concentrations of Hcy in a dose-dependent manner. Conclusions:Proliferation of vSMCs could be induced by Hcy, and the present investigation suggests that Hcy might contribute to the accelerated progression of atherosclerotic lesions with collagen alterations, which led to plaque instability and vulnerability.关键词
