Abstract
Objective To establish a HPLC-MS/MS method for deter-mination of nifedipine in human plasma.Methods After protein precip-itation, the plasma was separated on an Utimat AQ -C18 column, with a mobile phase of acetonitrile -20 mmol · L-1 ammonium acetate ( 58∶42 ) .The flow rate was 0.30 mL · min-1 and the column temperature 40 ℃.Quantification was performed in the negative ion multiple -reac-tion-monitoring ( MRM ) mode.Results Determination of nifedipine had good linearity in the concentration range of 0.03 -80 ng · mL-1.The lower limit of quantization was 0.03 ng · mL-1 , extraction recovery rate was from 94.71% to 102.3%, and the intra -day and inter -day RSD were both less than 9.8%.Conclusion This method is specific , simple, sensitive, rapid, accurate and suitable for determination of nife-dipine in human plasma.关键词
