氧化槐果碱诱导 SW480细胞凋亡的实验研究
Experimental study on the effects of oxysophocarpine inducting the apoptosis of cell lines SW480

Abstract
Objective To investigate oxysophocarpine inhibit prolifer-ation of human colon cancer cell line SW480 and induction of apopto-sis in which they are incurred , exploreing its mechanism at the same time.Methods Grouped according to the dose.MTT assay was used to detect the proliferation of SW480 cells.Fluorescence double stai-ning was used to observe changes in cell morphology and count the ra-tio of apoptotic cells.Flow cytometry ( FCM) were used to demonstrate the presence of apoptosis.Real -time PCR Detection were used to demonstrate changes in gene caspase-3 and Bcl-2.Results The experiment showed that OSC inhibited the growth of SW480 cells sig-nificantly.SW480 cells were induced to undergo apoptosis.Apoptotic gene caspase-3 mRNA was upregulated and Bcl-2 mRNA downreg-ulated. Conclusion Oxysophocarpine inhibits proliferation of SW480 cells in vitro, and arrest of cell cycle progression and apoptosis are involved in the mechanism.关键词
