Influence of ginkgo flavone aglycones on oxidized low-density lipoprotein induced inflammatory injury of endothelial cells

Abstract
Objective To observe the influence of ginkgo flavone agly-cones ( GA) on oxidized low-density lipoprotein ( ox-LDL) induced in-tercellular cell adhesion molecule 1 ( ICAM-1 ) , vascular cell adhesion molecule 1 ( VCAM-1 ) , monocyte chemotactic protein 1 ( MCP-1 ) in human aortic endothelial cells ( HAECs) and the mediated role of neclear factor-kappa B ( NF-κB).Methods Cultured HAECs in vitro were pretreated with different concentrations GA for one hour and then ox -LDL injury model of HAECs were established.MTT assay was used to detect endothelial cell activity, immunofluorescence was used to detect NF-κB activation, and enzyme-linked immunosorbent assay ( ELISA) was used to detect MCP-1, ICAM-1 and VCAM-1 level.Results GA 30-60 mg · L-1 pre-treatment group significantly inhibited ox-LDL-induced endothelial cells NF -κB activation ( P <0.05 ) and lowered the expression of VCAM -1 , ICAM -1 and MCP -1 ( P <0.05).Conclusion GA can effectively inhibit the expressions of ox-LDL -induced HAECs inflammatory factors, such as ICAM -1, VCAM-1 , and MCP-1.关键词
