摘要

Abstract

Objective To discuss the effect on Polygonum lowering streptozotocin ( STZ) rats′blood glucose and explore its possible mecha-nism.Methods The rats of type 1 of diabets( T1DM) was induced by 50 mg・ kg-1 STZ.Those rats were randomly divided into T1DM group, low-dose and high-dose group of Polygonum(1,2 g・ kg -1・ d -1 ), another nomal rats as the control group.T1 DM group and control group were gavaged equal 0.9%NaCl from the fourth week to the 12 nd week. The rats′weight and blood glucose would be measured in the fifth week and the twelve week. Then the pancreas tissue were made HE stai-ning.The level of insulin in the beta cells was detected by immunohisto-chemical staining,and apoptosis of islet cells was marked by terminal de-oxynucleotidyl transferase mediated dUTP biotin nick end labeling (TUNEL) .Results Compared with the control group , T1DM rats showed symptoms of polydipsia, polyuria, polyphagia, weight loss, high level of blood sugar and insulin structural damage.Compared with the T1DM group, high -dose group of Polygonum were generally better:weight was increasd ( P <0.05 ) , blood glucose was lower ( P <0.05 ) , while islet cells apoptosis index decreased significantly( P<0.05) ,insu-lin-positive expression was improved ( P <0.05 ) .Those indicators of low-dose group had no significant difference.Conclusion Polygonum&nbsp;can reduce the STZ rats′blood glucose probably by improving the expression of insulin and inhibiting the apoptosis of islet cells.
