Abstract
Objective:To investigate the possibility of the induction of muscle-derived stem cells(MDSCs)to differentiate into insulin-producing cells by islet and muscle-derived stem cells in neonatal rats(MDSCs)which were co-cultured in vitro.Methods:The primary cells of MDSCs in neonatal rats were extracted and identified,exogenous green fluorescent protein(GFP) were used to transfect MDSCs of the second generation.Rat pancreatic islets were extracted and then identified by dithizone,islets were co-cultured with MDSCs transfected by GFP.While only MDSCs were cultured as the control group.Results: Cell mass was found under a microscope on the fifth days of the co-culture group,and typical pancreatic insulin-secreting cell clusters was formed on the seventh day.Cell clusters identified by dithizone: a part of green cells was dyed scarlet,which indict that scarlet mass coming from MDSCs,but the same changes were not detected in the control group.Conclusion:MDSCs in neonatal rats can be induced into differentiate insulin-producing cells by co-culture MDSCs and islets.关键词
