古典H1N1亚型猪流感病毒反向遗传操作平台的建立
THE ESTABLISHMENT OF REVERSE GENETIC SYSTEM OF CLASSICAL H1N1 SWINE INFLUENZA VIRUS

Abstract
Eight gene segments of Influenza virus A/Swine/Guangdong/1/2011(H1N1) were amplified by RT-PCR, separately cloned into the vector of pBD and then transfected together into 293T cells. The supernatant of 293T cells was collected after 48 hours and inoculated into MDCK cells. When cytopathic effects(CPE) appeared in MDCK cells, virus titer was determined at 1:64 in hemagglutination test, which was comparable to the original wild strain. Eight genes of the rescued viruses were amplified in RT-PCR and also confirmed to be identical to the original virus. Therefore, rescued viruses were the same as the original influenza virus. The establishment of reverse genetic system of H1N1 subtype Swine influenza virus successfully, not only lay the foundation for exploring for the pathogenic mechanism, the infection mechanism and function, and so on, but also provide a basis for influenza vaccine of H1N1 swine influenza in the future.关键词
