Effects of Expression and Activity of SOD1 on Antagonistic Action of TMP on Gentamycin-induced Hearing Loss

摘要

Abstract

Objective To observe the effects of the expression and activity of Cu/Zn superoxide dismutase (Cu/Zn SOD or SOD1) on antagonistic action of TMP on Gentamycin-induced hearing loss.Methods 60 healthy guinea pigs were divided randomly into four groups: model group (GM group), therapy group (GM+TMP group), TMP group and normal control group, 15 in each group. GM group received intramuscular injection of Gentamycin sulfate 120 mg·kg-1·d-1 for 12 days. TMP group received TMP 30 mg·kg-1·d-1 intraperitoneal injection for 12 days. GM+TMP group received intramuscular injection of Gentamycin sulfate 120 mg·kg-1·d-1, intraperitoneal injection of TMP 30 mg·kg-1·d-1 for 12 days. The normal control group was administered with daily intramuscular injection of saline 2.5 ml·kg-1·d-1 for 12 days. Auditory brainstem responses (ABR) were used before and after the animals were tested,Western blot was used to test protein expression of SOD1 and ELISA to test SOD1 activity.Results Before treatment ABR thresholds in each group showed no significant difference (P>0.05);after treatment, GM group's ABR thresholds were significantly higher than GM+TMP group (P<0.01), TMP group and the normal control group's ABR thresholds before and after treatment did not change significantly (P>0.05). The protein expression of SOD1 increased significantly in GM+TMP group and GM group, and GM+TMP group was higher than that in GM group (P<0.05). The order of SOD1 activity in the cochlear tissue:GM+TMP group> normal control group≈ TMP group> GM group.Conclusion TMP can produce the anti-oxidant effects through adjust the protein expression and activity of SOD1.
