摘要

Abstract

Objective:To investigate the effect of human sperm hyaluronidase on male fertile ability. Method:44 semen samples of infertile patients were collected,and they were divided into 4 groups(A,B,C,D)according to the results of semen routine examine;26 samples were collected from normal fertile men as control group. The improved sodium hyaluronate - gelatin substrate film technique was used to determine the acrosomal hyaluronidase active intension,positive reaction rates and the enzyme reaction time in single sperm,the relationship between sperm morphology and hyaluronidase activity were studied;at the same time,acrosin activity was determined by fixed gelatin substrate film technique,and the correlationship of the two acrosomal enzymes was analyzed. CASA was made by sperm quality analyzer . Result:In the control group,the mean positive reaction rate of the sperm hyaluronidase was (70.84±19.20)%,and the mean diameters of hyaluronidase reaction area was(38.17±16.41)µm. The infertile groups were(37.01±35.13)%,(23.05±17.32)µm . In the experimental groups A,B,C and D,the mean positive reaction of hyaluronidase were(60.02±29.10)%,(16.11±4.64)%,(29.11±11.85)%and(53.04±28.60)%;the mean diameters of hyaluronidase reaction area were(26.92±17.21)µm,(6.90±2.00)µm,(8.22±3.37)µm and(31.40±18.60)µm. There were significant statistical difference in the positive reaction rate and active intension of the hyaluronidase between the fertile group and the infertile the group B,C(P<0.01). There were no significant statistical difference with the group A,D(P>0.05). There were obviously statistical correlationship between hyaluronidase activity and acrosin activity(r=0.810). Conclusion:The inactivity of sperm HYD is an important pathological reason of male infertility;detecting the activity of sperm HYD can be an clinical indicator to valuate the sperm function.
