Abstract
Objective:To investigate and evaluate the therapeutic effect of 188Re-tin colloid(TC)in animal models with malignant ascites.Method:188Re-TC colloid was prepared according to kit instruction,and labeling efficiency was determined by chromatography using radioactive thin-layer scanner.The animal models with malignant ascites were established through injecting 5×106/0.3 mL sarcoma180 cells,cultured in vitro,into abdominal cavity of ICR mice.The experimental animal models were divided into 3 different dose-groups after 3 day of culture,that were 37,18.5 and 9.25 MBq/mL of 188Re-TC respectively.The control groups were also divided into 3 groups,12 mice in each group,which included(1)the mice of malignant ascites by intraperitoneal injection of saline;(2)the normal ICR mice by intraperitoneal injection of 1 mCi/mL 188Re-TC;(3)the normal ICR mice without any treatment.Different collection times of intraperitoneal radioactive distribution images in mice were captured by gamma camera at 3,24 h and 48 h post injection.Then peritoneal effusion was collected to detect the expression of Bax protein by western blotting method,while the major abdominal organs were also collected to evaluate histopathological changes.After injection of 188Re-TC,the body weight and survival time of mice were monitored every two days and observed 80 days in all. Statistical analysis was performed with SPSS(version 10.7).Survival analysis was calculated by Kaplan Meir statistical methods,difference among groups was compared by log Rank method.Result:Labeling efficiency of the 188Re-TC was higher than 99%.Homogeneous distribution of radioactivity in the abdomen was found in 3,24 and 48 h images of treated mice without abdominal intraperitoneal leakage.Dose-dependent expression of Bax protein was confirmed.Histopathological analysis showed that mice treated with 188Re-TC,compared to the ones treated with saline,expressed more in the liver cell degeneration,spleen white pulp atrophy,neutrophil infiltration,intestinal cell adhension,degradation of gastric glands,and so on.Two groups of mice injected with 1 mCi/mL 188Re-TC showed a trend to loss body weight at the beginning,compared with the other groups,but then the body weight of them gradually increased after 8-10 days, and there was no significant difference between groups(P>0.05).Survival analysis results in 80 days after 188Re-TC injection showed that the survival time of 37,18.5 and 9.25 MBq/mL group were(59.0±8.5)d,(63.6±7.1)d and (54.8±7.0)d respectively,and were significantly increased compared with the saline group [(20.0±1.9)d](P<0.001). Conclusion:188Re-TC not only has the better physical properties,but also holds the good curative effects on malignant ascites with less side-effects,which may provide more choice for malignant pleural effusion and ascites.关键词
