Abstract
Objective To explore the mechanisma of LiCl which plays a role in protecting cells through regulating the activity of GSK-3β. Method Plasmids pBACE1-mychis and pAPPswe were transfected into SHSY5Y cells by LipofectaminTM2000. The cells were treated with LiCl and then were collected to determine the protein levels of GSK-3β,β-catenin and CyclinD1by Western blot.MTS, Brdu detect cell proliferation. Result Western blot results showed that in the cells, LiCl inhibited the expression of GSK-3β, and promoted the expression ofβ-catenin, CyclinD1. LiCl signiifcantly inhibited the growth of cells transfected. SHSY5Y. MTS inhibition rate was 23.45%, and Brdu inhibition rate was 22.58%. Conclusion GSK-3βis a potential target for treating with AD. LiCl can prevent AD through inhibiting the activity of GSK-3β.关键词
