Abstract
Objective To establish and compare the HPLC and UV method to detect the content of ponatinib raw material drug. Methods In the HPLC method,the mobile phase was acetonitrile - 0. 2% triethylamine buffer solution(85 : 15),the Agilent Zor-bax Extend C18 analytical column(250 mm × 4. 6 mm,5 μm)was adopted,the flow rate was 1. 0 mL / min,the temperature was 30 ℃ , the injection volume was 20 μL and the detection wavelength was 280 nm. In the UV method,the sample was dissolved in anhydrous ethanol and detected at the wavelength of 280nm. Results In HPLC method,the linear range of ponatinib was 1 - 40 μg / mL( r =0. 999 9),the average recovery rate was 100. 00% with RSD = 0. 27%( n = 9). In the UV method,the linear range of ponatinib was 0. 5 - 20 μg / mL( r = 0. 999 9),the average recovery rate was 99. 29% with RSD = 0. 89%( n = 9). The relative error in the UV method was 1. 432 - 6. 102 times of that in the HPLC method. Conclusion The HPLC method is more accurate in the detection results than the UV method,which is more suitable for the content analysis of ponatinib raw material drug.关键词
