Abstract
Objective To study the effect of N-trimethyl chitosan(TMC)used as a transdermal enhancer with different degrees of quat-ernization(DQ) on the in vitro properties of celecoxib liposomes(CLXL) in order to provide the basis for screening better TMC. Methods CLXL was prepared by using the film dispersion method. TMC(TMC20,TMC40 and TMC60) with various DQ (20%,40%and 60%) was mixed with CLXL,the ratio of TMC amount and CLXL was fixed as 1:4( W/ W). The morphology,size,Zeta potential and entrapment efficiency of CLXL after mixing with TMC were investigated. With azone as the positive control and CLXL without any enhancer as the negative control,the cumulated amount and the rate for penetrating mouse in vitro skin were studied by using Franz diffusion cell method. Results TMC and DQ showed no significant effect on the morphology and the entrapment efficiency of CLXL, while the particle size of CLXL was increased to some extent and the Zeta potential was changed from negative to positive after TMC mixing,furthermore,the changes showed positive correlation with DQ of TMC;compared with the negative control,various TMC exhibited significant transdermal absorption promoting effect,moreover the promoting effect was more significant with the DQ increase. Among the three kinds of TMC,TMC60 showed no statistically significant difference compared with the positive control group. Conclusion TMC with different DQ has certain effect on the in vitro properties of CLXL,in which,the transdermal absorption promoting effect of TMC60 is similar to that of the same amount of azone,which is worthy of being further studied.关键词
