Abstract
Objective:To establish a HPLC method for the content determination of puerarin in gegen wendan granules.Methods:Samples were determined by HPLC on the C18 column(250 mm×4.6 mm,5μm). The column temperature was at 30 ℃;the inject volume was 10μL. The mobile phase was consisted of methanol-0.2% phosphoric acid(18∶82). The flow rate was 1.0 mL/min at UV detection wavelength of 250 nm. The number of theoretical plate calculated by puerarin peak should be not less than 3 000.Results:There was a good linear relationship of puerarin within the range of 0.020 02 ~1.000 84μg,r= 0.999 9. The average recovery of puerarin was 97.45%,RSD = 1.62%(n= 6).Conclusion:The method is convenient and accurate with a good specificity and reproducibility,it is suitable for the content determination of puerarin in gegen wendan granules.关键词
